MS, HPLC and LC-MS answer related but different questions. The right route starts with the decision you need to make, not with the instrument name.
Mass spectrometry (MS)
MS is suited to checking an observed molecular mass against a theoretical value. It supports identity but does not provide a complete purity profile on its own.
HPLC analysis
HPLC separates detectable components and is commonly used to report main-peak area percentage. It shows chromatographic purity under the selected conditions but does not by itself establish the molecular identity of every peak.
LC-MS analysis
LC-MS connects chromatographic separation with mass detection. It can help investigate complex samples, co-eluting components or peaks that require mass assignment. The added information generally means a higher cost and a more project-specific method.
A simple decision guide
Define whether the question is identity, purity, content, safety or impurity investigation. One sample may need more than one result.
- Identity only: consider MS
- Purity only: consider HPLC
- Basic identity and purity: request MS plus HPLC
- Complex mixture or peak investigation: discuss LC-MS
- Amount of active peptide: request a validated or justified quantitative method
