HPLC purity describes the relative chromatographic profile. Net peptide content estimates the actual amount or proportion of peptide after accounting for materials such as water and counter-ions. These results should not be used interchangeably.

Two different denominators

An HPLC area percentage compares detector responses among included peaks. Net content relates peptide amount to the sample, vial or stated mass. Because the denominators are different, the numerical values may differ even when both tests are correctly performed.

Why powder weight can mislead

Lyophilized material can contain residual moisture, counter-ions and other non-peptide material. In formulated products, declared excipients may add further mass. Weighing the powder alone therefore does not establish the active peptide amount.

What a useful report states

A quantitative result should identify the method, calculation basis, unit and relevant sample preparation. Results such as mg/vial, mg/g or percentage are not equivalent and must be read with their unit.

How to request the right test

Tell the laboratory whether you need chromatographic purity, active amount per vial, concentration in a liquid or another quantitative claim. Supplying the expected range and formulation helps the laboratory propose an appropriate method.
